PENENTUAN KADAR TANIN TOTAL PADA EKSTRAK ETANOL DAUN JERUJU (Acanthus ilicifolius L.) MENGGUNAKAN METODE SPEKTROFOTOMETRI UV-VIS
Abstract
Jeruju (Acanthus ilicifolius L.) is a medicinal plant known to contain various secondary metabolites such as triterpenoids, saponins, alkaloids, phenolics, flavonoids, and tannins, which contribute to its pharmacological activities, including antioxidant, antimicrobial, and anti-inflammatory effects. This study aimed to determine the total tannin content in the ethanol extract of Jeruju leaves using UV-Visible spectrophotometry. The extraction was carried out using the maceration method with 96% ethanol as the solvent. Qualitative analysis using FeCl₃, potassium ferricyanide, and ammonia resulted in characteristic color changes, indicating the presence of tannins. Quantitative analysis was performed using a UV-Vis spectrophotometer at a maximum wavelength of 759 nm, with gallic acid as the reference standard for the calibration curve. The results showed that the total tannin content in the ethanol extract of Jeruju leaves was 76.72 mg GAE/g extract. These findings suggest that Jeruju leaves are a promising natural source of tannins and may have potential for further development in traditional herbal medicine.
